How it works

Lyse in SDS. Trap the protein. Wash the SDS away. Digest in place.

Solubilise

Lyse in a strong SDS buffer, which recovers membrane proteins, FFPE material and lipid-rich tissue that milder detergents leave behind. Reduce and alkylate in the same tube.

Trap

Acidify and dilute into a methanolic binding buffer: the protein forms a fine particulate suspension that is captured in the quartz depth filter. SDS, salts and contaminants wash through.

Digest and elute

Protease goes directly into the trap. The protein is held in a large, accessible surface, so digestion typically completes in about an hour. Peptides elute ready for desalting or injection.

S-Trap™ is particularly effective for plasma, FFPE tissue, low-input samples and high-lipid matrices where in-solution protocols lose material or suffer ion suppression.

Formalin-fixed blocks need hot SDS to reverse cross-links; the trap removes that SDS before digestion, so decades-old material becomes tractable.

Do you supply individual solutions?

Yes, Solutions 1 to 7, the reductant/alkylator pack and Si-Trap™ Solution L are all stocked for laboratories that already own columns or plates.

What is Tryp-N™ for?

It cleaves N-terminal to lysine and arginine, mirroring trypsin, so peptides carry the basic residue at the N-terminus. The b-ion-rich spectra are complementary to tryptic data, useful for PTM localisation, sequence coverage and electron-based fragmentation (Huesgen et al., Nat Methods 2015).

What if setting up the workflow in-house is not practical for us?
